Rufus Sewell's character Major Henry Nicholls is asking the angel Castiel to prove that heroes are possible. If angels really were pure and incorruptible, as Major Nicholls believes, then he would be able to trust Castiel without a doubt. However, as we know from season five of Supernatural, many angels have been corrupted by their own personal agendas. There also appears to be a problem with the purity of heaven itself, as seen in episode sixteen of season one, titled All Hell Breaks Loose: Part One.
